Can someone explain the difference and what would be required to use an L32 (series III supercharged) fuel rail on a 2002 L67 pontiac bonneville? I read somewhere that I would need to add a pressure regulator that is on the L67 fuel rail. What else is required? Do you block of the return fuel line do you connect them so they are in a loop? I don't have the knowledge to understand what the differences are and why they made the change from return to returnless on the newer models.

A stock return system pressurizes the system between the pump and the FPR (passes injectors). The FPR bleeds off any excess through the return line to the tank.

A return-less system pressurizes the system in the same way but varies the pressure via the pump and doesn't return. So, there's only one port on the fuel rail vs. 2 for a return system.

What you can do if you want to use the L32 fuel rail is plumb the stock L67 feed line from the tank into a 3 port aftermarket FPR. The FPR will have a pressurized outlet that goes to the returnless rail and the 3rd port will bleed off excess pressure to the return line. I ran a setup like this in my i30 for like 8 years.
